Spray it OUTSIDE!!!! No one has mentioned that yet and you really do NOT want to be breathing it in. I spray outside (either on the porch or the patio), by the time I close the door and walk back upstairs to my loft where I usually do my work, it's dry. And no one answered your first question... YES you do want to do a sealer coat first! This will help keep the face up materials from staining head so most everything can be removed should you change your mind/sell the head. It doesn't have to be thicker or anything, same light mist. How you use the sealer doesn't change whether you are using pastels, watercolor/pencils, or acrylic.
I don't use acrylic, but I imagine you want to let it dry completely before spraying your next coat of MSC. It's a good idea to do things in layers, and spray in between, that way if you really like what you did on the eyes and then mess up the eyebrows, you don't ruin what you like while trying to remove what you don't.
